Cross- linking of surface- bound IgGs leads to the adhesion of neutrophils to endothelial cells. HUVECs were grown in 96 well plates until confluence. Neutrophils were incubated with calcein and with or without FcγRs blocking antibodies (A) or anti-CD18 antibody [37] (10 μg/ml) (B) before addition of PPP, as described in Methods. Neutrophils were then added to endothelial cells in the presence of anti-human IgG antibodies for 30 minutes. Non adherent neutrophils were discarded and the amount of adherent cells was quantified as the amount of fluorescence bound to endothelial cells. Results are expressed as mean +/- SEM of three independent experiments. * = P < 0.05, ** = P < 0.005, *** = P < 0.0005 (Mann–Whitney test).
