FACS analysis of DC phenotypes Human peripheral blood mononuclear cells (PBMCs) were isolated from the whole blood of normal donors. PBMC monocytes were purified in complete culture medium for 90 min. The remaining adherent cells were induced by 800 U/ ml recombinant GM-CSF and 1000 U/ ml IL-4 for 5 days. Cells were cultured for 2 days in the presence of 1000 U/ ml TNF-α to induce final maturation. The cells were immunostained with FITC and anti-CD11c, CD83 and CD86 antibodies, and the phenotypes were analyzed by flow cytometry. These mature dendritic cells express high levels of CD11c, CD83 and CD86. Three repeated experiments showed similar results
