Effect of TLR-2 neutralizing antibody on the induction of MMP-9 expression by HKLM. THP-1 cells were treated anti-huTLR-2-IgA (neutralizing mAb; Nab; 2ug/ml) or isotype-matched control antibody (IgA) (2ug/ml) for 30 minutes before the addition of HKLM. Cells and cell culture supernatants were collected at 24 hr after HKLM treatment. MMP-9 mRNA was quantified by real-time PCR (A) and secreted levels of MMP-9 were measured in the supernatants by ELISA (B). SEAP reporter activity (degree of AP-1/NF-κB activation) was determined in the supernatants as described in the material methods (C). Data are shown from three independent experiments. Data are presented as mean ± SE. An asterisk (*) represents P-value of <0.05.
