SR141716A decreases TNF-a secretion and expression in LPS-stimulated mature adipocytes. Panel A: The concentrations of TNF-a in the medium of mature adipocyte cultures, treated or not with LPS 1 μg/mL alone or in combination with SR141716A were measured at 6 hours by ELISA. SR141716A was used at 50, 100, 200 and 400 nM. Results are expressed in pg/mL. The graph shows the mean ± SD of the results from 3 patients (n = 6 for each condition, for each patient). ***P < 0.001 and *P < 0.05, versus LPS-treated cells. Panel B: TNF-a gene expression was determined at 4 hours of treatment in mature adipocyte cultures, treated or not with LPS 1 μg/mL alone or in combination with 200 nM SR141716A.
